The involvement of specific t cells in the pathogenesis of metamizole-induced agranulocytosis

چکیده
Background Non-chemotherapy related, drug-induced agranulocytosis is a rare idiosyncratic reaction, which may be fatal. Along with betalactames, the analgesic metamizole is reported to occasionally cause agranulocytosis. The disease results in a severe reduction of granulocytes rendering affected patients susceptible to bacterial and fungal infections. The pathogenesis of drug-induced agranulocytosis is complex as non-immune (mainly toxic) and immune mechanisms might be involved.

GM‐CSF as successful salvage therapy of metamizole (dipyrone)‐induced agranulocytosis with Fournier's gangrene and severe septic shock in an adolescent
This case report describes the successful use of granulocyte and macrophage colony-stimulating factor as salvage therapy and an alternative to hematopoietic stem cell transplantation in a 14-year-old adolescent with metamizole (dipyrone)-induced agranulocytosis and severe septic shock.
متن کاملMetamizole (Dipyrone) as an Alternative Agent in Postoperative Analgesia in Patients with Contraindications for Nonsteroidal Anti-Inflammatory Drugs.
PURPOSE Nonsteroidal anti-inflammatory drugs (NSAIDs) play an important role in multimodal pain management. In patients with a contraindication for NSAIDs, pain management is challenging. A recent Dutch anesthesiology guideline propagates the use of metamizole (dipyrone) in these patients. Metamizole is a controversial drug, its use being previously discouraged because of the risk for agranuloc...

Background and Objectives: Multiple sclerosis (MS) is an autoimmune neurodegenerative disease of the central nervous system (CNS). Although, the contribution of various cells such as B cells, CD8+ T cells, microglia/macrophages, dendritic cells, asterocytes and mast cells in the pathogenesis of MS have been demonstrated, however, it seems that autoreactive myelin specific CD4+ T cells pla...
متن کاملPossible Involvement of a Specific Cell Surface Receptor for Calprotectin-Induced Apoptosis in Colon Adenocarcinoma and Carcinam Cell Lines (SW742 and HT29/219)
Calprotectin, a calcium-bound protein complex, is abundant in the cytosol of neutrophils. It has been reported that this protein has an apoptotic activity in tumor cells. Since calprotectin increases in colorectal cancer, this study was conducted to investigate, for the first time, the cytotoxicity/apoptotic effect of calprotectin on HT29/219 and SW742 colon carcinoma and adenocarcinoma cell li...
متن کاملPharmacological characteristics of metamizole.
Metamizole (dipyrone) is a popular analgetic, non-opioid drug, commonly used in human and veterinary medicine. In some cases, this agent is still incorrectly classified as a non-steroidal anti-inflammatory drug (NSAID). Metamizole is a pro-drug, which spontaneously breaks down after oral administration to structurally related pyrazolone compounds. Apart from its analgesic effect, the medication...
